Product Overview
The DYKDDDDK peptide is a widely used epitope tag for fusion protein expression, detection, and purification. It can be incorporated at the N-terminus, C-terminus, or within the sequence of a recombinant protein.
Because the DYKDDDDK peptide is highly hydrophilic, it is readily exposed on the surface of many fusion proteins, helping facilitate antibody recognition. When positioned at the N-terminus, the tag can also be cleaved by Enterokinase (EK protease) at the C-terminal side of its aspartic acid residue.
Anti DYKDDDDK Antibody-HRP combines specific recognition of the DYKDDDDK tag with Horseradish Peroxidase (HRP) conjugation, providing a convenient format for direct detection of tagged proteins in Western blot assays.

HRP Chemiluminescent Detection Principle
The antibody is conjugated to Horseradish Peroxidase (HRP). During chemiluminescent detection, HRP catalyzes the reaction of luminol with hydrogen peroxide.
The reaction generates an unstable peroxide intermediate that decomposes through an electronically excited state. As the excited species returns to its ground state, light is emitted, allowing the DYKDDDDK-tagged fusion protein to be visualized on a Western blot.
About the DYKDDDDK Tag
DYKDDDDK is commonly used as an epitope tag in recombinant protein expression and purification. The tag can be positioned at different locations within a fusion protein, including the N-terminus and C-terminus.
Its hydrophilic nature can facilitate surface exposure and antibody recognition. When placed at the N-terminus, the sequence can also be cleaved by Enterokinase, providing a route for removal of the tag from the target protein.
