Product Overview

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a key enzyme involved in glycolysis. It catalyzes the oxidation and phosphorylation of glyceraldehyde-3-phosphate, producing 1,3-bisphosphoglycerate.

GAPDH is widely distributed across tissues and is commonly used as a reference protein in molecular biology experiments because of its generally abundant expression. It is frequently used as an internal reference in applications such as Western blot (WB) and qPCR.

Anti GAPDH Antibody-HRP is produced by conjugating Horseradish Peroxidase (HRP) to a GAPDH antibody. The HRP conjugate enables direct chemiluminescent detection of GAPDH during Western blot analysis.

HRP Chemiluminescent Detection

During Western blot detection, luminol and hydrogen peroxide are used as chemiluminescent substrates. In the presence of HRP, luminol reacts with hydrogen peroxide to generate an unstable peroxide intermediate.

The intermediate rapidly decomposes through an electronically excited state. When the excited species returns to its ground state, light is emitted, allowing visualization of the GAPDH reference protein.

Western Blot Workflow

STEP 01
Protein Separation

Separate proteins by electrophoresis.

STEP 02
Membrane Transfer

Transfer separated proteins onto a membrane.

STEP 03
GAPDH Detection

Apply the HRP-conjugated GAPDH antibody.

STEP 04
Chemiluminescence

Add chemiluminescent substrate and visualize the signal.
