Product Overview
The BCA (bicinchoninic acid) assay is a widely used colorimetric method for measuring protein concentration. The assay is based on the reduction of copper ions by protein peptide bonds under alkaline conditions, followed by the formation of a colored complex between bicinchoninic acid and copper ions.
During the reaction, copper divalent ions (Cu²⁺) are reduced to copper monovalent ions (Cu⁺). Two BCA molecules then complex with Cu⁺ to form a purple-colored complex with strong absorbance at 562 nm.
Within the appropriate working range, the absorbance of the resulting complex is related to protein concentration, allowing protein samples to be quantified using a suitable spectrophotometer or microplate reader.
BCA Assay Principle
Protein peptide bonds reduce Cu²⁺ to Cu⁺ under alkaline conditions.
BCA molecules complex with Cu⁺ to generate a purple-colored complex.
The resulting complex is measured at 562 nm for protein quantification.
