Product Description

Supreme TC Grade Agar is a plant tissue culture solidifying agent used to provide essential physical support for the growth and development of plant explants in culture media.

Agar is a polysaccharide derived from seaweed that forms a gel-like matrix when incorporated into culture media. This solid structure supports plant cells and tissues while allowing nutrients, hormones, and other essential components to remain distributed throughout the medium.

Agar and gellan gum are among the most commonly used solidifying agents for plant tissue culture applications.

Assay Protocol

The following protocol describes preparation of 1 liter of tissue culture medium using Supreme TC Grade Agar:

1. Add 800 mL of distilled water to a beaker.
2. Add 4.54 g of PCT's MS Media, or an alternative medium appropriate for the plant species.
3. Add 30 g of sugar and PPM™ (generally 1–2 mL/L).
4. Make up the volume to 1000 mL.
5. Adjust the pH to approximately 5.4–5.8.
6. Add 6–8 g of Supreme Tissue Culture Grade Agar while stirring.
7. Autoclave the prepared medium for 20 minutes at 15 psi and 121°C.
